چکیده
The association between obesity and osteoarthritis (OA) in joints not subjected to mechanical overload, together with the relationship OA metabolic syndrome, suggests that there are systemic factors related disorders involved phenotype of OA. aim this work is study effects palmitate oleate on cellular metabolism an “ vitro ” model human chondrocytes. TC28a2 chondrocyte cell line was used analyze effect mitochondrial glycolytic function, Adenosine triphosphate (ATP) production lipid droplets accumulation. Palmitate, but oleate, produces dysfunction observed a lower coupling efficiency, maximal respiration spare respiratory capacity. Glycolytic function showed rates both capacity reserve when cells were incubated fatty acids (FAs). rate total ATP values chondrocytes palmitic acid (PA). formation increased FA conditions, being significantly higher oleic (OL). These results may help explain, at least part, close pathologies OA, as well elucidate some can define

Oleate Prevents Palmitate-Induced Atrophy via Modulation of Mitochondrial ROS Production in Skeletal Myotubes
Accumulation of saturated fatty acids contributes to lipotoxicity-related insulin resistance and atrophy in skeletal muscle. Conversely, unsaturated fatty acids like docosahexaenoic acid were proven to preserve muscle mass. However, it is not known if the most common unsaturated oleate will protect skeletal myotubes against palmitate-mediated atrophy, and its specific mechanism remains to be el...
متن کاملOleate protects against palmitate-induced insulin resistance in L6 myotubes.
Oleate has been shown to protect against palmitate-induced insulin resistance. The present study investigates mechanisms involved in the interaction between oleate and palmitate on insulin-stimulated glucose uptake by L6 skeletal muscle cells. L6 myotubes were cultured for 6 h with palmitate or oleate alone, and combinations of palmitate with oleate, with and without phosphatidylinositol 3-kina...
متن کاملACE inhibition prevents myocardial infarction-induced skeletal muscle mitochondrial dysfunction.
Heart failure is associated with alterations in cardiac and skeletal muscle energy metabolism resulting in a generalized myopathy. We investigated the molecular and cellular effects of angiotensin-converting enzyme inhibition (ACEi) on skeletal muscle metabolism in infarcted animals. Myocardial infarction (MI) was obtained by left descending coronary artery ligation. Sham, MI, and MI-treated ra...
متن کاملAdipocyte Fatty Acid-Binding Protein Promotes Palmitate-Induced Mitochondrial Dysfunction and Apoptosis in Macrophages
A high level of circulating free fatty acids (FFAs) is known to be an important trigger for macrophage apoptosis during the development of atherosclerosis. However, the underlying mechanism by which FFAs result in macrophage apoptosis is not well understood. In cultured human macrophage Thp-1 cells, we showed that palmitate (PA), the most abundant FFA in circulation, induced excessive reactive ...
متن کاملMitochondrial Dysfunction Prevents Repolarization of Inflammatory Macrophages.
Macrophages are innate immune cells that adopt diverse activation states in response to their microenvironment. Editing macrophage activation to dampen inflammatory diseases by promoting the repolarization of inflammatory (M1) macrophages to anti-inflammatory (M2) macrophages is of high interest. Here, we find that mouse and human M1 macrophages fail to convert into M2 cells upon IL-4 exposure ...
